Occupational Summary

Perform a variety of complex technical duties involved in conducting laboratory tests, experiments and determinations in support of Duke University research operations.

Work Performed

· Setting up and operating laboratory equipment required for specific experiments; and preparing and maintaining solutions and other reagents for laboratory operations. Carrying out experiments using a variety of standard molecular and cell biology studies and histology approaches including, but not limited to, protein and nucleic acid purification, quantification, and analysis; plasmid DNA generation, manipulation, and analysis; bacteria and mammalian cell culture and analysis; mouse and human tissue harvest and analysis using histological approaches and molecular biology assays. Maintaining a laboratory notebook with accurate and timely documentation of laboratory work; meeting with the laboratory PI, as needed, to review data and progress and plan future research experimental priorities. Where appropriate, preparing or assisting with preparation of publication-quality figures or written reports for presentation or scientific publication.

· Assisting with the management of the laboratory mouse colony, including maintaining colony census and record of animal use; obtaining tissue for experimental analyses, including PCR-based genotyping.

· Assisting with maintaining day-to-day laboratory operations, including monitoring and replenishing supplies and reagents; cleaning and maintaining laboratory equipment; and assisting PI in preparing documents for institutional reporting, such as in maintaining IRB or IACUC protocols or biological/chemical safety protocols.

· Reviewing work of less experienced laboratory personnel, such as undergraduate students, within designated areas, and assist in training new personnel.

Required Qualifications at this Level

Education/Training: Work generally requires a bachelor's degree in botany, biology, zoology, psychology or other directly related scientific field.

Skills: Experience from work or laboratory-based educational courses in pertinent molecular and/or cell biology assays, such as general biology/chemistry knowledge. A combination of some or all of the following: nucleic acid and protein extraction and measurement; plasmid DNA preparation, manipulation, and analysis; polymerase chain reaction (PCR); agarose and/or SDS-polyacrylamide gel electrophoresis and blotting; mammalian and/or bacterial cell culture and analysis; fluorescence and light microscopy; experience with personal computer hardware and software for word processing, data storage and analysis, digital image processing, and scientific literature searches. Familiarity with molecular biology, skeletal muscle development, and/or cancer biology are preferred.
